ABSTRACT

Viscum plants,the evergreen perennial parasitic shrubs or subshrubs,are mainly distributed in tropical and subtropical regions. There are about 70 Viscum species around the world,including 11 species and one variety in China. Mistletoe lectins are typeⅡ ribosome-inactivating proteins( RIPs) extracted from Viscum plants with anticancer and immunoregulatory activities. Many studies have focused on the mistletoe lectins isolated from V. album in Europe and V. album var. coloratum distributed in South Korea,respectively,and several preparations,such as Iscucin Ⓡ,were developed and clinically applied for cancer treatment. Although Viscum plants are widely distributed in China,only a few studies of mistletoe lectins have been reported. The recent progress of mistletoe lectins was reviewed from extraction,purification,quantitative/qualitative detection,molecular structure,pharmacological activities,toxicities,and clinical application,aiming at providing a reference for in-depth research and utilization of mistletoe lectins produced in China.

ABSTRACT

There are 16 species and 7 varieties of medicinal plants of Trollius in China, with effects in clearing heat, detoxification and swelling, they are used for the treatment of acute and chronic tonsillitis, acute otitis media, chronic bronchitis and urinary tract infections. Through retrieval of literatures on nasturtium from 1972 to 2019 in a number of databases, such as CNKI, Wanfang Database, Baidu Academic and PubMed, the cultivation and quality control methods, extraction and purification processes, chemical composition, pharmacological effects, pharmacokinetics and metabolism of medicinal plants of the genus nasturtium were summarized. The researches of cultivation mainly focus on the establishment of seed treatment and tissue culture system. Quality control is mainly based on orientin and flavonoids. Research on extraction and purification technology mainly focused on the extraction and purification of total flavonoids. In terms of chemical composition, there are currently more than 100 major compounds isolated from this genus, including flavonoids, organic acids, alkaloids, coumarins and styrenes. Among them, there are 80 flavonoids, 12 organic acid monomers, 3 alkaloid monomers, 4 coumarin monomers and 14 styrene monomers. In terms of pharmacological action, there are mainly in vitro experiments of crude extracts of medicinal materials. The data of pharmacokinetics and metabolism mainly focus on the study of flavone monomers.

ABSTRACT

Chinese medicine polysaccharide is an important active biological macromolecule, which has a broad application prospect. However, there are still many deficiencies in the quality evaluation and control of polysaccharides. Based on the existing problems in the quality control of traditional Chinese medicine polysaccharides, current review analyzes the methods of extraction, separation and purification, characteristic identification, content determination and structure analysis of Chinese medicine polysaccharides, and draws the following conclusions: ①Based on the clinical application of Chinese medicine, decoction is recommended as the extraction method in the basic study of effective substances of Chinese medicine polysaccharides; ②On the basis of impurity removal, HPGPC can achieve the separation, purification and content determination of Chinese medicine polysaccharides at the same time, supplemented by MS or NMR can achieve the quantitative and qualitative analysis of Chinese medicine polysaccharides; ③Based on the characteristic identification, select the suitable pure polysaccharide, dextran or monosaccharide reference; ④HPSEC-MALLS-RID is specific, accurate, and beneficial to the study of structure-activity mechanism of polysaccharides. The review suggested that the comprehensive evaluation and control of the quality of Chinese medicine polysaccharides should be based on extraction, separation and purification, on the premise of characteristic identification, on the basis of content determination, supplemented by structural analysis, which is to provide useful reference for the quality research of Chinese medicine polysaccharides.

ABSTRACT

Lycii Fructus is a precious traditional medicinal and edible herb with multifunction, in which polysaccharide is believed to be a predominant active substance with multifarious biological functions and broad utilization prospect. In this review, the new technology application in extraction, purification, and structural elucidation of Lycii Fructus polysaccharide was systematically untangled, the current research situation and development trend were outlined, and the potential product groups and industry prospect in Lycii Fructus polysaccharide industrialization were analyzed to provide scientific reference for the enhancement of effective utilization and industrial chain of Lycii Fructus resources.

ABSTRACT

The study of spectrum-effect relationship is one effective method according to the pharmacodynamic material basis and mechanism of action of Chinese materia medica (CMM). The first step is to adopt different modern analysis methods to obtain the compound information of CMM, secondly choose proper animal model to evaluate the pharmacological activity, and then combine two kinds of information mentioned above to find out the main effective substance based on the results of statistical methods. Nowadays, the study of spectrum-effect relationship is widely used in many different aspects. In this article, three different applications of spectrum-effect relationship were mainly summarized, including the study of the quality assessment of CMM, the study of extraction and purification method of CMM and the study of processed CMM, in order to provide reference for the further study of CMM.

ABSTRACT

Objective: To extract and purify thepolysaccharide from Portulacaoleracea and evaluate its anti-oxidant activity. Methods: Orthogonal experiment design L9(43) was employed to optimize the traditional extraction process. The processes of deproteinization and depigmentation were also investigated. Specifications of membrane with different apertures was used to purify the crude polysaccharide. The anti-oxidant activity of the polysaccharide was also evaluated by Fenton reaction and autoxidation of pyrogallol. Results: The polysaccharide was extracted at 100℃ for 3h with four times repeated to give the highest extraction rate. The recycling rates of Sevage method, thicloroacetic acid and hydrochloric acid method to remove protein were 17.05%, 9.66%, and 16.61%, respectively. The recycling rates of active carbon, H2O2, acetone-anhydrous ethanol, and chloroformn butanol to remove pigment were 17.04%, 17.09%, 27.06%, and 17.58%, respectively. Much of the polysaccharide were detected in the liquid penetrated from the membrane with apertures of 0.8μm.The clearance rate of polysaccharidescavenge hydroxyl radical and superoxide radicalwere 58.89%, and 38.89%. Conclusion: The preferred aperture of microfiltration and ultrafiltration membrane can purified P.oleracea. Polysaccharide antioxidant experiment indicates that the polysaccharide could effectively scavenge hydroxyl radical and superoxide radical with dose dependent relationship.

ABSTRACT

Objective: A novel one-step extraction and purification method by using superparamagnetic adsorbents was established for separation of tanshinones from Salvia miltiorrhiza. Methods: Firstly, superparamagnetic poly hydroxyethyl methacrylate (PHEMA) microspheres were prepared by dispersion polymerization method, and their adsorptive performances of tanshinones were investigated, then these microspheres for purification of tanshinones were coupled into extraction process of tanshinones. The feasibility and separation efficiency of this one-step method were further studied. Results: The prepared PHEMA microspheres with the average diameters of 1.2 μm had superparamagnetic property and good adsorption performance for tanshinones. In comparison with two steps method including extraction and purification, one-step method showed significantly higher extraction efficiency of three tanshinones from 0.179, 0.093, and 0.452 mg/g in 5 h to 0.279, 0.176, and 0.575 mg/g in 0.5 h, with less separation process, greatly shorter time consumption from 5.5 h to 0.5 h, and good purification results. Conclusion: One-step method proposed is demonstrated feasible and has the merits of high extraction efficiency, less separation process, good purification, and rapid separation speed.

The quality controls are performed by near infra-red fingerprint spectrum. HPLC. X-ray diffraction Fourier spectrum, and ITS2 barcode. The nutrients of E. ferox contain large amount of amino acids, fatty acids and various microelements; medicinal ingredients are identified to be polyphenol, sesquineolignan. tocopherol, cerebroside, and cyclic dipeptide. The pharmacological effects of E. ferox extracts include antioxidant, free radical scavenging, hypoglycemic activity, decreasing urine protein, bacteriostasis and prevention of gastric mucosal injury. Treatment with E. ferox preparations for diabetes and chronic nephritis are performed in clinics. This review summarizes the study of E. ferox in recent years for further research and development.

ABSTRACT

Objective To develop an efficient method to isolate and purify the main components isoaloeresin D and aloin from Aloe vera for its industrial production. Methods High-speed counter-current chromatography was used to isolate isoaloeresin D and aloin in a one-step separation from dried crude extract ofA. vera. The biphasic solvent system the lipophilic phase was selected as the mobile phase and the apparatus was rotated at 840 r/min. The effluent was detected at 254 nm. Results Isoaloeresin D (53.1 mg) and aloin (106.9 mg) were separated from the crude extract (384.7 mg) with the purities of 98.6% and 99.5%, respectively. Conclusion HSCCC is a powerful technique for isolation and separation of chemical composition from aloe.

ABSTRACT

Objective To clarify the influence on component and pharmacological action of Astragalus polysaccharides (APS) as complementary therapeutic agents prepared by different extraction and purification techniques. Methods Components of APS prepared by different extraction and purification techniques were analyzed, and these APS were used for synergy and attenuation of chemotherapy, radiotherapy treatment with H22 liver cancer and Lewis lung cancer of tumor-bearing mice, and also used for the regulation of immune function to immunosuppression mice. Results Experimental data were analyzed by means of statistical method to get pharmaco-result: A3 (extracted by microwave assistance and purified by membrane separation) > A4 (extracted by refluxing and purified by membrane separation) > A1 (extracted by refluxing and no purification)≈ A2 (extracted by microwave assistance and no purification). There were no significant differences on pharmacodynamic action between A1 and A2. However, compared with A1 and A2,it was worth noting that A3 and A4 exhibited good pharmacodynamic action. Then A3-in and A4-in, the samples in dialyzer after dialysis, were separated and purified to get homogeneous APS, which were the principal constituents of APS in dialyzer, with the molecular weight (Mw) of 7669 and 14 142 determined by HPGPC, respectively. The average Mw of APS outside of the dialyzer, A3-out was 3102 and A4-out 3256, which were the main compositions of A3 and A4, accounted for 79.63% and 53.92%, respectively. Conclusion APS with Mw about 5000 Da exhibit better antitumor effect and immunological activity. Refluxing, microwave assistance extractions, and membrane enrichment techniques bring different cases on Mw distribution, components and pharmacodynamic action, and obviously exhibit relationship among component, Mw distribution, and pharmacological action.

ABSTRACT

Six DNA extraction methods and four DNA purification methods were compared and analyzed in this study to get higher quality DNA from the rhizospheric soil of Fritillaria thunbergii Miq.Results showed that higher purity DNA were harvested by pretreating the soil with 20 mmol/L EDTA(pH 7.5),then isolating soil DNA with CTAB-SDS-frozen-thawing,and further purified by agarose method.The recovery rate of this soil DNA was about 44.00 ?g/g ? 2.65 ?g/g soil,and they were qualified for the microbial diversity analysis in the rhizospheric soil of F.thunbergii Miq based on the 16S rDNA sequence.

ABSTRACT

OBJECTIVE:To extract and purify organic acids with macroporous resins from Folium Isatidis.METHODS:Folium Isatidis was extracted with 70% aqueous ethanol(pH=2)by Soxhlet extraction.The process was optimized by employing orthogonal test design with the content of anthranilic acid as the index.RESULTS:The resin HPD100 had much better adsorption capacity and desorption ratio by using 3 BV 60% aqueous ethanol to extract organic acids in Folium Isatidis than other resins.The results showed that HPD100 possessed the better ecomic under the concentration of sample 0.18mg?mL-1,the condition of pH 3.5,and flowing rate of 2BV?h-1.CONCLUSION:Macroporous resin HPD100 may be value of isolating and purifying the organic acids from Folium Isatidis.

ABSTRACT

Objective To study the method of extraction,purification,and analysis of total flavonoids of Radix Glycyrrhizae(TFG).Methods The extracting effects of three different ways(ultrasonic wave,heating recirculation,and maceration)for TFG were compared.TFG was refined with resin absorption and the effects of different parameters on yield and purity were observed.Naringin scanning method was used to analyze TFG.Results TFG extracting percentage is 5.46 %by ultrasonic wave method,6.12 %by heating recirculation,and 5.04 %by maceration,the effect of heating recirculation being the best.In the test of resin absorption of TFG,the heating recirculation composition is mainly located in 40 %~60 %ethanol elution parts.Conclusion The best method for extracting TFG is heating recirculation in above three methods,Elution with 80%ethanol continuously after water elution is the the optimal resin purification method for TFG.
